Background:
High-calorie, obesogenic diets are strongly associated to accelerated aging and foster the emergence of typical aging-associated disease like metabolic syndrome and cardiovascular complications. Data from animal studies suggest that disturbed polyamine homeostasis, specifically insufficient polyamine supply in specific tissues, could be causally involved in the negative effects of obesogenic diets. Polyamines are essential for cell-autonomous function, e.g., autophagy, as well as non-autonomous processes, e.g., (sterile) inflammation. Yet, despite these critical roles of polyamines, the molecular etiology of polyamine disbalance under high-calorie diets is insufficiently understood.
Hypothesis and Objectives:
We hypothesize that insufficient spermidine supply and/or maladaptive changes in polyamine metabolism is a causal driver for accelerated aging under high-calorie, obesogenic diets and that spermidine supplementation may ameliorate these effects. Through time-resolved molecular fingerprinting of relevant animal tissues, we will pin down the impact of high-calorie diets on polyamine homeostasis. These results will be complemented by clinical biopsies to evaluate the translational potential of the findings. Importantly, diversity of gut microbiota, an important source of bioavailable polyamines, will be closely monitored.
Methodology:
As part of the MetAGE Cluster of Excellence, you will work hands-on with rodents and investigate transcriptomic, proteomic and metabolomic changes of various tissues, supported by an in-house data science platform. Microbiome analyses will be conducted in close collaboration with the Medical University of Graz. The MetAGE consortium will provide access to human samples, allowing the translation of preclinical findings into human aging research.
We offer an annual gross salary of € 52,007.20 for a fulltime position.
