The aim of the laboratory is to decipher the relationship between the structure of the genes and their function and between genotype and phenotype, from gene to patient. To carry out these projects, Unit 1078 relies on genetics and molecular genomics approaches, genetic epidemiological data, expertise in transcription, splicing and the development of original research models such as the baker's yeast. The expected results are: an increasing knowledge concerning the molecular basis of the gene of interest and also the development of new diagnostic and therapeutical strategies in some model diseases.
The stroke team is composed of a PhD student, a post-doc skilled in stroke experimental models lead by a senior scientist (S.T). This team is embedded in the larger research group of Emmanuel Genin on functional genetics with wide competence from molecular biology, biochemistry, genetic epidemiology. ST is a Professor of Neurology specialized in stroke and develops since many years research on neuroprotection (see references). He is at the head of the Neurology Department at the CHRU of Brest. His team collaborates with molecular biologists, geneticans and bioinformaticians. The aim of this translational team is focused on ischemic stroke to develop therapeutic drugs in ischemic stroke. Patents have already been filed patents in the field.
Mission (scientific project)
Stroke represents the third cause of death in industrialized countries and the second cause in the world according to the world health organization (WHO). Cerebral edema increases intra-cranial-pressure and is the main cause of death after a stroke at the acute phase. There is no medical therapeutic treatment of cerebral edema. The only proposed treatment is neurosurgical, hemicraniectomy. Our previous work showed that a CDK (Cyclin Dependant Kinase) inhibitor was able to decrease brain edema of 37% after focal stroke ischemia compared to controls. The main objective of this project is to study the effect of a CDK inhibitor on cerebral edema in a rat cerebral focal ischemia model. Our previous work clearly showed that a CDK inhibitor had a pleiotropic benefic effect including a decrease of brain edema, neuroprotective properties by decreasing the size of the infarction, and an improvement of motor deficit. This new project will allow better understanding of the effect of CDK inhibitors on brain edema and may contribute to the development of a new therapeutic strategy in stroke. A patent on the use of a CDK inhibitor on stroke has been already delivered. Further study on CDK inhibitor and brain edema in stroke will allow to better understand the mechanism of brain edema decrease and finally to propose new therapeutic strategies for the treatment of brain edema which represents the major cause of acute death after stroke.
Doctor (PhD) in Life Sciences preferentially in Neurosciences, with experience on cell culture with a maximum 3 years of experience after thesis defense (1). Experience in tissue analysis (histological staining, immuno-histochemistry), cellular and molecular biology (FACS, RT-PCR) is required.
